Abstract

Mesophyll protoplasts isolated either from axenic shoot cultures of Hyoscyamus albus L. and H. muticus L. or from suspension cultures of H. muticus undergo sustained divisions in modified NT and DPD medium with division frequencies up to 50 %. In H. muticus morphogenesis could be readily induced even when small calluses (3–4 weeks old) were directly transferred from the original protoplast culture medium onto solid differentiation medium. In most cases morphogenesis was via shoot and root formation but when MS–medium containing activated charcoal was used, the formation of embryo–like structures was observed. In H. albus, in contrast, it has as yet only been possible to induce root formation in protoplast–derived cultures.
