Antiapoptotic Effects of Propolis Extract and Propol on Human Macrophages Exposed to Minimally Modified Low Density Lipoprotein

Summary

An aqueous extract of propolis and the phenolic component of propolis, propol, were assayed for antioxidative and antiapoptotic properties. Both additions inhibited Cu2+-initiated low density lipoprotein (LDL) oxidation as characterized by a reduction of the lag time, reduced the increase of relative electrophoretic mobility during oxidation and markedly diminished apoptosis of human macrophages exposed to minimally modified (mmLDL). Moreover, aqueous propolis extract and propol blocked the mmLDL-induced decrease of glutathione (GSH) and the activation of the transcription factor NF-kB in these cells. The potent phenolic antioxidant propol thus expands the capability of cells to neutralize oxidative stress and to prevent apoptosis and is therefore suggested to significantly contribute to the antiinflammatory and antioxidative effects of propolis.

Zusammenfassung

Antiapoptotische Effekte von Propolisextrakt und Propol auf die durch minimal-modifiziertes Low Density Lipoprotein induzierte Apoptose in humanen Makrophagen

Ein wäßriger Extrakt von Propolis und des phenolischen Inhaltsstoffs Propol wurden auf antioxidative und antiapoptotische Eigenschaften untersucht. Der wäßrige Extrakt und Propol inhibierten die Cu2+-vermittelte Low Density Lipoprotein (LDL)-Oxidation, was sich über eine Verlängerung der lag-time manifestierte, reduzierten die oxidationsbedingte Zunahme der relativen Mobilität in der Elektrophorese und verringerten die durch minimal modifiziertes LDL (mmLDL) in Humanmakrophagen ausgelöste Apoptose. Darüber hinaus wurde sowohl durch den wäßrigen Extrakt als auch durch Propol die durch mmLDL verursachte Reduktion der Glutathionkonzentration und die Aktivierung des Transkriptionsfaktors NF-kB verhindert. Das phenolische Antioxidans Propol verbessert auf diese Weise die Fähigkeit von Zellen, oxidativen Stress zu kompensieren und Apoptose zu verhindern: es sollte deshalb einen deutlichen Anteil an den Propolis zugeschriebenen antioxidativen und anti-inflammatorischen Eigenschaften haben.
