Hyaluronidase, Elastase and Matrix Metalloproteinase-1 Inhibitory Activity of Standardized Extract of Piper betel Linn. Leaf

The experiment was designed to screen anti-wrinkle (skin) activity of P. betel L. through in-vitro hyaluronidase, elastase and matrix metalloproteinase-1 (MMP-1) inhibition. The P. betel leaf was extracted with methanol and fractionated with ethyl acetate, n-butanol and water. Enzymatic assays were performed with extract and fractions (1.56 – 50 µg/mL) along with standard oleanolic acid [1]. Lead compounds were isolated from the bioactive fractions, quantified through RP-HPLC and characterized by GC-MS, 1H NMR and IR spectroscopic studies. The methanol extract showed significant (P < 0.01) anti-hyaluronidase and anti-elastase activity with IC50 of 3.21 ± 0.40 µg/mL and 16.06 ± 0.54 µg/mL respectively and good MMP-1 inhibition compared to oleanolic acid. The isolated compounds hydroxychavicol and chlorogenic acid were also showed similar enzymes inhibition. The RP-HPLC analysis revealed the amount of hydroxychavicol (28.56% w/w) and chlorogenic acid (0.4% w/w). P. betel showed potent inhibitory activity against hyaluronidase, elastase and MMP-1. Therefore, it can have a potential as an anti-wrinkle agent.
