Assessment of Helichrysum sp. extracts on in vitro keratinocyte proliferation and differentiation: potential use of plants for improved wound healing

Helichrysum italicum (Roth) G. Don fil. and Helichrysum stoechas (L.) Moench are species of the Asteraceae family with wide distribution in the Mediterranean basin. Helichrysum species have been long known as a medicinal plant in Europe for treatment of various medical conditions for at least 2,000 years [1]. In ethnopharmacological surveys Helichrysum sp., mostly H. italicum, is described for treatment of respiratory and digestive diseases, for skin inflammatory conditions and improved wound healing, as well as an antimicrobial use and analgesic remedy [2].

Phytochemical composition of Helichrysum genus includes phenolic compounds (chlorogenic-, caffeic acid, quercetin glucoside, etc.), monoterpenes (neryl acetate, -propanoate, α-pinene, etc.), sesquiterpenes (γ-curcumene, β-selinene, α-selinene) and others. Hydroethanolic extracts (7:3 v/v) of the two species were studied under in vitro conditions for potential effects on skin cells, using HaCaT cell line and primary human keratinocytes. Metabolic activity, proliferation and cellular differentiation were assessed by MTT assay, BrdU ELISA, and Western Blot analysis for differentiation-specific cytokeratin 10 (CK10) and involucrin (INV).

Both extracts significantly stimulated cell proliferation of HaCaT cells at concentrations of 10 µg/mL after 24h and 48h. Incubation of primary keratinocytes for 7 days with 10 µg/mL of the extracts triggered cells in the differentiation state, as monitored by increased CK10 expression, while INV expression stayed more or less unchanged. No influence on cellular vitality was observed at 10 µg/mL while concentrations of ≥100 µg/mL exerted toxic effects.

These results confirm the traditional use of Helichrysum sp. for improved wound healing.
