Zusammenfassung

Die Positronen-Emissionstomografie bietet in Abhängigkeit von den verwendeten Liganden ein hohes Maß an Strukturspezifität bei gleichzeitiger räumlicher Information unter in-vivo Bedingungen. Dennoch fand sich neben den durchaus etablierten [18F]FDG-PET Messungen kaum eine fundierte Anwendung mit direkter therapeutischer Konsequenz. Fortschritte in der Pharmakotherapie psychischer Erkrankungen können die PET allerdings auch klinisch rasch in eine relevante Schlüsselposition bringen. Am Beispiel der in Erprobung befindlichen Immun- Therapeutika für die Alzheimer-Demenz-Erkrankung wird gezeigt, dass die Liganden- Entwicklung zur Frühdiagnose (Amyloid-Imaging) integraler Bestandteil des gesamten Therapie- Prozesses werden kann. Die Entwicklung von geeigneten Amyloid-bindenden Liganden nimmt daher aktuell einen breiten Raum der Neuro-PET-Forschung ein.

Summary

Molecular imaging by positron emission tomographie (PET) provides valid estimates for the distribution and extent of some structures in the central nervous system. Depending on the choice of radioligands its outcome can be of high structural specifity. Whereas [18F]FDG is a commonly used radiotracer, other ligands are not routinely used in clinical practice. Recent advantages in the treatment of Alzheimer’s disease however require highly sensitive as well as specific tools for the detection of early disease states. Amyloid imaging therefore emerged to be one of the major fields of interest in radiochemistry. The development of suitable and handy amyloid-binding ligands appears to be an important requirement for the applicability of innovative therapy strategies of Alzheimer’s disease.
