Introduction:

The study's goal was to discover differences in leukocyte subsets between head and neck cancer patients (HNC-P) and healthy adults (HA) from a population-based cohort.

Material and Methods:

10-color flow cytometry (FCM) was used for in-depth immunophenotyping of peripheral blood (PB) samples of 608 HA and 101 therapy-naïve HNC-P. Absolute cell count (AbsCC) of leukocyte (CD45+) subsets were determined.

Results:

Comparing 608 HA and 101 HNC-P, differences (p < 0.01) in AbsCC were detected in granulocytes (side scatter high, SSChigh), neutrophiles (SSChighCD16+), monocytes (SSCmedCD14high), classical monocytes (SSCmedCD14highCD16dim), lymphocytes (SSClow), T-lymphocytes (CD3+CD16-CD56-), and T-cell subpopulations (CD3+CD8+ cytotoxic T-cells, CD3+CD4+ T-helper cells, and CD3+CD4+CD8+ double-positives).

Conclusion:

AbsCC allow for discrimination of HA and HNC-P but, however, had low power to discriminate HNC-P and HA. Consequently, we are aiming on analyses of relative cell counts to facilitate this approach.
