Introduction Nanoparticles (NP) as drug-carrier are in the spotlight of recent research. Possible applications reach from the modulation of anti-inflammatory genes up to targeted tumor-therapy. The focus of this work is on the biologic effects of calcium phosphate (CaP) nanoparticles on the cytokine secretion of epithelial and mesenchymal cells. Methods: CaP-NP with and without conjugated Cetuximab were applied on epithelial tumor cells (FaDu) as well as on mesenchymal stem cells (MSC) originating from the nasal mucosa. Modulation of the cytokine secretion was observed using enzyme linked immunosorbent assays (ELISA) for interleukin-6, -8 and macrophage migration inhibitory factor following stimulation with TNF-alpha and INF-gamma. Cell-targeting of cetuximab-conjugated CaP was observed using an co-culture assay.

Results The exposition with CaP-NP showed concentration- as well as time-depended effects in both cell types. Both showed an increase of the stimulatory effect of cytokines as well as increased secretion of IL-6, -8 and MIF. Rising the concentration of NP led to an increase in cytokine secretion. Co-culture showed no cell-selective accumulation of cetuximab conjugated NP.

Discussion CaP-NP modulate the interleukin-secretion of epithelial as well as mesenchymal cells of the head and neck region. The effect of interleukins in the tumor environment is controversial. Before therapeutic use detailed analysis of those effects, focusing on the involved immune cells, is essential.
