The herbal preparation, STW 5-II, consisting of hydro-alcoholic extracts of Iberis amara, Matricaria recutita, Carum carvi, Melissa officinalis, Mentha × piperita and Glycyrrhiza glabra, was studied on ulcerative colitis induced in rats by feeding them with 5% dextran sodium sulphate in drinking water for 1 week. STW 5-II (2 ml/kg) was administered orally to rats for 1 week before DSS and for a further week after. After 24 hours, the rats were sacrificed. The colon length and weight were measured, and colon homogenates were prepared so as to assess various mediators for antioxidant and anti-inflammatory activity. STW 5-II guarded to a large extent against changes in colon length and mass index (oedema formation). Furthermore, it protected against changes in myeloperoxidase, reduced glutathione, glutathione peroxidase and superoxide dismutase induced by DSS indicating good antioxidant activity as well as against changes in TNF-α, indicating good anti-inflammatory activity as well. The results could open a new field of therapeutic usefulness for this herbal gastroprotective agent.
