Significance
Fmoc/t-Bu SPPS depends on harsh, TFA-driven cleavage, creating side reactions and sustainability
issues. The authors’ developed Fmoc/Pic platform replaces t-Bu with a photocleavable, feedstock-derived Pic group, enabling acid-free, on-resin
peptide release under mild conditions, thus offering a sustainable and operationally
simple alternative for complex peptide synthesis.
Comment
Fmoc/Pic chemistry enables visible-light cleavage of stable Pic protecting groups,
replacing TFA-dependent steps in SPPS. This sustainable platform simplifies peptide
synthesis and expands access to complex peptide structures.
