Lokale Faktor-XIII-Bestimmung in der Dickdarmmukosa

Zusammenfassung

Faktor-XIII-Aktivität wurde in endoskopisch gewonnenen, homogenisierten Biopsieproben von Patienten mit Colitis ulcerosa und Morbus Crohn mittels eines Amin-Inkorporationstests bestimmt. Lösliches Dinitrophenylcadaverin (DNPC) wird von aktiviertem Faktor XIII an Casein gekoppelt, das wiederum an die Wandung von Microtiterplatten adsorbiert ist. Das gebundene DNPC wird mittels spezifischem Antikörper detektiert. Es fand sich eine Abhängigkeit der Faktor-XIII-Aktivität von der histologischen Klassifikation (nicht entzündet, leichte Entzündung, schwere Entzündung) mit deutlich niedrigeren Werten bei schwerer Entzündung. Die beschriebene Methode eignet sich zur funktioneilen Messung von Faktor XIII und anderen Proteintransglutaminasen in Gewebe- und Zellhomogenisaten und anderen trüben Probenmaterialien.
