Hintergrund

Der NO-sGC-cGMP-Signaltransduktionsweg (NO = Stickstoffmonoxid; sGC = lösliche Guanylatcyclase; cGMP = zyklisches Guanosinmonophosphat) ist bei vielen Herzkreislauferkrankungen einschließlich der pulmonal-arteriellen Hypertonie (PAH) gestört. Bei Riociguat (BAY 63 – 2521) handelt es sich um einen sGC-Stimulator, der den cGMP-Spiegel sowohl in Synergie mit NO als auch unabhängig davon erhöht. Ziel dieser Studie war es, die Rolle der NO-sGC-cGMP-Signalgebung in einem Modell mit schwerer PAH zu untersuchen und die Wirkung der sGC-Stimulation durch Riociguat sowie der PDE5-Inhibition durch Sildenafil auf die pulmonale Hämodynamik und das vaskuläre Remodeling bei schwerer experimenteller PAH zu bewerten.
