ZUsammenfassung

Bereits früher publizierte Daten zeigten günstige Effekte von Thymian bei entzündlichen Prozessen. Um diese Erkenntnisse zu vertiefen, wurde in verschiedenen experimentellen Modellen ein Thymianextrakt auf die hier zugrundeliegenden molekularbiologischen Mechanismen in vitro untersucht. Nachgewiesen wurde eine Hemmung der LPS-induzierten Zytokin-Freisetzung von IL-1β, PGE2 sowie TNF-α in humanen Monozyten, eine Hemmung der LPS-induzierten Phosphorylierung und eine Aktivierung von IκBα sowie eine Inhibition der Transkriptionsaktivität von NF-κB und AP-1 in der Bronchialepithel-Zelllinie A549. Ein weiterer interessanter Befund war eine antagonistische Aktivität des untersuchten Thymianextraktes am TRPA1-Rezeptor, ein Rezeptor, der Hustenreize vermitteln kann. Die Anwendung des Thymianextraktes bei Erkältungs- bzw. Bronchitis-bedingtem Husten, bei dem entzündliche Prozesse auf bzw. in der Schleimhaut involviert sind, darf daher als plausibel gelten.

Abstract

Molecular basis of anti-inflammatory effects of thyme extract, Thymus vulgaris L. and Thymus zygis L., herba

Previously published data showed beneficial effects of thyme in inflammatory processes. In order to deepen these findings, an in vitro investigation of thyme extract was carried out in various experimental models for the underlying molecular-biological mechanisms. These studies demonstrated an inhibition of LPS-induced cytokine release IL-1β, PGE2 and TNFα in human monocytes; an inhibition of LPS-induced phosphorylation and activation of IκBα as well as inhibition of transcriptional activity of NF-κB and AP-1 in the bronchial epithelial cell line A549. An additional interesting finding was the antagonistic activity of the examined thyme extract at the TRPA1 receptor, a receptor that can mediate cough stimuli. The use of thyme extract for cough caused by colds or bronchitis, in which inflammatory processes are involved on or in the mucous membrane, can therefore be considered plausible.
