Zusammenfassung

Die Proliferation von Blastzellen und die Kinetik der makromolekularen RNS- und DNS-Synthese unstimulierter und mit Phytohämagglutinin stimulierter Lymphozyten von Gesunden und Patienten mit Morbus Hodgkin wurden in vitro untersucht. Die Nucleinsäuren wurden von mit 3H-Thymidin und 3H-Cytidin inkubierten Zellen durch Perchlorsäure extrahiert. Es fand sich kein Unterschied im Stoffwechsel unstimulierter Lymphozyten von Gesunden und von Patienten mit Lymphogranulomatose. Während normale Lymphozyten eine maximale Reaktion zwei bis drei Tage nach der Exposition mit Phytohämagglutinin aufwiesen, zeigten Hodgkin-Lymphozyten eine verzögerte Reaktion mit einem Maximum nach fünf Tagen. Bei Hodgkin-Lymphozyten war zusätzlich eine Frühreaktion der Nucleinsäuresynthese nachzuweisen, abhängig von dem Stadium der Erkrankung und der Lymphozytenzahl im peripheren Blut. Die Ergebnisse liefern Hinweise auf eine normale und eine abnorme Lymphozytenpopulation bei Morbus Hodgkin.

Summary

The proliferation of blast cells and the kinetics of macromolecular RNA and DNA synthesis of unstimulated and phytohaemagglutininstimulated lymphocytes was investigated in normal subjects and patients with Hodgkin's disease. Nucleic acid was extracted with perchloric acid from cells incubated with 3H-thymidine and 3H-cytidine. The metabolism of unstimulated lymphocytes was the same in patients with Hodgkin's disease and those in normal persons. Normal lymphocytes had a maximal reaction two to three days after exposure to phytohaemagglutinin, while lymphocytes from patients with Hodgkin's disease had a delayed response with a maximum after five days. In addition, lymphocytes from Hodgkin's disease had an early reaction of nucleic acid synthesis, which depended on the stage of the disease and the number of lymphocytes in peripheral blood. The results suggest that there is both a normal and abnormal lymphocyte population in patients with Hodgkin's disease.

Resumen

Reacción retardada de los linfocitos en la enfermedad de Hodgkin a la fitohemaglutinina

La proliferación de blastocitos y la cinética de la síntesis macromolecular del ARN y ADN de los linfocitos, estimulados con fitohemaglutinina y sin estimular, de sanos y enfermos con morbo de Hodgkin fueron exploradas. Los ácidos nucleínicos se extrajeron por ácido perclórico de las células incubadas con timidina-3H y citidina-3H. No se encontró ninguna diferencia en el metabolismo de los linfocitos no estimulados de sanos y enfermos con linfogranulomatosis. Mientras que los linfocitos normales mostraron una reacción máxima de dos a tres días después de la exposición con fitohemaglutinina, mostraron los linfocitos del Hodgkin una reacción retardada con un máximo después del quinto día. En los linfocitos de la enfermedad Hodgkin se pudo comprobar, además, una reacción precoz de la síntesis del ácido nucleínico, dependiente del estadio de la afección y del número de linfocitos de la sangre periférica. Los resultados suministran indicios de una populación linfocitaria anormal y normal en la enfermedad de Hodgkin.
