Durch die Einführung zielgerichteter anti-tumoraler Therapien und der damit einhergehenden Identifikation neuer prädiktiver sowie prognostischer molekularer Marker konnte auch im Bereich der gastrointestinalen Tumoren eine Individualisierung der medikamentösen Therapiekonzepte erreicht werden. Neben klinischen Faktoren, Behandlungsziel, Patientenwunsch und genetischen Polymorphismen kommt der molekularen Stratifizierung der Patienten für die individuelle Behandlung eine zunehmend bedeutende Rolle zu.

Primärer Ansatz einer zielgerichteten Therapie ist der präzise Angriff auf die Tumorzelle, verbunden mit geringen Nebenwirkungen und einer verbesserten Gesamtverträglichkeit. Während zytotoxische Chemotherapeutika vor allem in den Prozess der Zellteilung eingreifen, interferieren zielgerichtete anti-tumorale Medikamente mit spezifischen Molekülen, die eine entscheidende Rolle in der Signaltransduktion der Zelle bei Tumorwachstum und -progression spielen.
