Nasal microbiome of patients with granulomatosis with polyangiitis

Introduction:

Inflammatory diseases are linked to a dysbiosis of the local microbiome. The objective of this study was to determine whether there is a specific composition of the nasal microbiome of patients with granulomatosis with polyangiitis (GPA).

Methods:

63 nasal swabs of 26 patients with GPA, 21 patients with rheumatoid arthritis (RA) and 27 control patients (CPs) were taken in the Department of ENT, head and neck surgery of the Christian- Albrechts- University of Kiel and the Department of Rheumatology of the University of Lübeck. The swabs were processed for standard microbial analysis using 16S rRNA amplicon sequencing to survey the composition of the nasal microbiome. Due to frequent detection of Staphylococcus aureus (S. aureus) in recent findings a specific diagnostic polymerase chain reaction (PCR) for these organisms was initiated as well.

Results:

The 16S rRNA analysis did not show any statistically significant difference in both the alpha- and beta-diversity between all patient groups. The PCR detected a higher rate of S. aureus in GPA patients (15/19) compared to CPs (4/23, p < 0.0005) and RA patients (9/25; p = 0.0046).

Conclusions:

The PCR analysis suggest a specific local microbiome with increased existence of S. aureus in patients with GPA compared to the control groups.
