Etwa 5–20 % aller Mamma- und Ovarialkarzinome werden auf genetische Aberrationen zurückgeführt. Bei BC machen BRCA1/2-Mutationen mindestens 25 % des familiären Risikos aus. Dies führte zu der Annahme komplexerer, polygenetischer Muster. Bislang sind für Mamma- und Ovarialkarzinome insgesamt 11 Suszeptibilitätsgene charakterisiert. Die Studie mit insgesamt 15 404 Teilnehmerinnen bestätigte bekannte Befunde, ergab neue Konstellationen und stellte Risikogene in Frage.
