Establishment of Raman spectroscopy for investigation of laryngeal leukoplakia

Introduction:

The pre- and intraoperative determination of dignity of laryngeal leukoplakia is challenging.

Raman spectra are dependent on the molecular composition of the analyzed tissue.

The aim of this study was to evaluate the Raman spectra of dysplastic, malignant and healthy tissue of the glottis.

Material and methods:

The investigation of dysplastic, malignant and healthy tissue of the glottis was performed by using the BioRam System of CellTool. Therefore, epithelial and subepithelial stroma cells were stimulated with a 785nm diode laser under microscopical control. In this pilot study, deparaffined tissue slices were analyzed. The „The Unscrambler“ X 10.3 Software was used for multivariate data analysis.

Results:

Specific averaging spectra were identified for healthy, dysplastic and malignant tissue. In the principal component analysis, the main differences were observed in the 1st and 2nd principal component. The averaging spectra differentiated primarily at 1350 cm-1, 900 cm-1 and 800 cm-1.

Discussion:

Our data show promising results. The biological information assessed by Raman spectroscopy could be useful to differentiate between healthy, dysplastic and malignant tissue of the glottis. Since the laser only scans the tissue but does not destroy biological matrix, the method may be used for in vivo evaluations. Thus, possible future applications for Raman spectroscopy could be the pre- or intraoperative determination of the dignity and the evaluation of resection margins in patients with laryngeal leukoplakia.
