Generation of a cell-rich SVF gel for regenerative therapy approaches with the VoiceInject system
Introduction The so-called "stromal vascular fraction (SVF) gel" is a mechanically processed product of adipose tissue consisting of extracellular matrix and mesenchymal stromal cells (ASC). In the present work, the extraction of SVF-gel using the VoiceInject system was investigated for use in regenerative therapeutic approaches in the head and neck region.
Material and methods Fat tissue specimens resected during body lift procedures was used to harvest adipose tissue using the liposuction cannula of the VoiceInject system. After the processing steps recommended by the manufacturer, a uniform emulsion was prepared from the fat tissue and centrifuged again. Afterwards, the SVF gel appeared as a middle phase between oil and aqueous solution. The cells contained in the SVF gel were cultured and characterised in detail with regard to proliferation behaviour, differentiation capacities and stem cell properties.
Results A cell supension corresponding to an adipocyte-poor cell fraction in the extracellular matrix was obtained from the harvested adipose tissue. The contained cells showed the morphology typical for ASC and a characteristic proliferation behaviour. Furthermore, their osteogenic, adipogenic and chondrogenic differentiation potential could be confirmed.
Conclusion The SVF gel contains ASC with their typical characteristics and is thus a promising injection material for regenerative therapeutic approaches. Compared to pure SVF, the combination with the extracellular matrix should enhance the positive effect of the cells. An application in chronic inflammatory diseases, e.g. of the lacrimal gland or salivary glands, is the aim of further investigations and represents a new therapeutic option.
