Seit der Erstbeschreibung [18] der Angiotensin-II-(AngII)-induzierten arteriellen Hypertonie vor über 40 Jahren hat die Erforschung des Renin-Angiotensin-Aldosteron-Systems (RAAS) wesentlich zum Verständnis der Pathogenese kardiovaskulärer Erkrankungen beigetragen. Angiotensin-Konversionsenzym-Hemmer (ACE-Hemmer) und AngII-Typ1-Rezeptor Blocker (AT1-Rezeptor-Antagonisten) gehören heute zum Standard der Behandlung der arteriellen Hypertonie, der Herzinsuffizienz und der diabetischen Nephropathie. Zahlreiche Studien haben belegt, dass die Prognose kardiovaskulärer Erkrankungen durch medikamentöse Intervention am RAAS deutlich verbessert wird [3] [11]. Neben der blutdrucksteigernden Wirkung von AngII sind wachstumsfördernde Effekte auf Zielzellen des kardiovaskulären Systems, wie z. B. Kardiomyozyten, kardiale Fibroblasten und glatte Gefäßmuskelzellen pathogenetisch von zentraler Bedeutung. In der folgenden Übersichtsarbeit sollen zunächst die therapeutischen Interventionsmöglichkeiten am RAAS diskutiert werden und am Beispiel der mitogen a ktivierten Proteinkinasen (MAP-Kinasen) Signaltransduktionsmechanismen aufgezeigt werden, die für AngII-induzierte Wachstumsprozesse relevant sind.
