The aim of this study was to evaluate the effect of three different desensitizers (Gluma® Desensitizer, Admira Protect, Hyposen) on the de- and remineralization of root dentine in vitro.

The root surfaces of 40 freshly extracted human third molars were coated with an acid-resistant nail varnish exposing two rectangular windows on the root surface. All teeth were randomly assigned into 4 groups: K: untreated control; G: treated with Gluma® Desensitizer; A: treated with Admira Protect; H: treated with Hyposen. All specimens were demineralized for 14 days with acidified gel (HEC, pH 4.8, 37 °C). Before remineralization using a NaF containing calcium-phosphate buffer solution one half of each window was covered. From each window a dentinal slab was cut. The depth of the demineralized areas was determined using a polarized light microscope.

In the desensitizer-groups the lesion depths were significantly reduced compared to the untreated control. The comparison between de- and remineralized groups showed a significant reduction of lesion depth in all cases (p<0.05, Tukey's test).

It can be concluded that the demineralization of the root surface can be reduced by the application of desensitizing agents. However, the remineralization was limited by the used sealants compared to the control.
