Complexing of Reduced Technetium and Tin (II) by Chelating Phosphate Compounds

Complexing of reduced 99mTc, "Tc and of 113Sn(II) by pyrophosphate was studied, using the in vivo distribution in the rat as an indicator for complex formation. 99mTc-pyrophosphate was only formed at very low technetium concentrations, otherwise colloid formation occurred. Reduced 99mTc in trace amounts and 113Sn(II) complexed equimolarly with pyrophosphate were concentrated in bone and excreted in urine in comparable amounts. The in vivo distribution of 32P- orthophosphate and -pyrophosphate was characterized by a considerable uptake in the liver and a poor urinary excretion. The distribution patterns of 32P-pyrophosphate, complexed equimolarly with tin(II), remained unchanged. Despite these findings it is concluded that real chelates are formed, since non-complexed reduced 99mTc was not concentrated in bone. Two variants of reduced technetium could be discerned: a 99mTc-kidney agent and a 99mTc protein-bound agent. Some evidence was found that reduced technetium may change its oxidation state, but it seems much more likely that both agents are technetium(IV) compounds. It is suggested that the kidney agent is hydrated technetium dioxide whereas the protein- bound agent is a charged technetium(IV) compound. Only hydrated technetium dioxide is obviously complexed by chelating phosphate compounds.

Die Komplexbildung zwischen reduziertem 99mTc, 99Tc und 113Sn(II) mit Pyrophosphat wurde an Hand der in vivo Verteilung in der Ratte als Indikator für die Komplexbildung untersucht. Lediglich bei sehr niedrigen Technetiumkonzentrationen entstand 99mTc-Pyrophosphat, ansonsten bildete sich Kolloid. Als Komplex mit Pyrophosphat wurden 99mTc in Spurenmengen und 113Sn(II) in gleichen molaren Konzentrationen in vergleichbaren Mengen im Knochen angereichert und mit dem Urin ausgeschieden. Die in vivo Verteilung von 32P-Orthophosphat und -Pyrophosphat war durch eine beträchtliche Anreicherung in der Leber und durch eine geringe Ausscheidung durch den Urin gekennzeichnet. Das Verteilungsmuster von 32P-Pyrophosphat blieb trotz gleicher molarer Komplexbindung mit Zinn(II) unverändert. Trotzdem wird die Schlußfolgerung gezogen, daß reelle Chelate gebildet werden, da nicht als Komplex vorliegendes reduziertes 99mTc keine Knochenanreicherung zeigte. Zwei Varianten von reduziertem Technetium konnten unterschieden werden: eine nierensuchende und eine proteingebundene 99mTc Verbindung. Es fanden sich einige Anhaltspunkte dafür, daß reduziertes Technetium seine Wertigkeit ändern könnte. Es scheint jedoch wahrscheinlicher, daß in beiden Fällen Technetium(IV) Verbindungen vorliegen. Es wird angenommen, daß es sich bei der nierensuchenden Verbindung um Technetiumdioxydhydrat und bei der proteingebundenen Verbindung um eine geladene Technetium(IV) Verbindung handelt. Lediglich Technetiumdioxydhydrat geht offensichtlich eine Komplexbindung mit chelatbildenden Phosphatverbindungen ein.
