Preclinical analyses of the cell-assisted lipotransfer (CAL) for injection laryngoplasty

    The enrichment of adipose tissue with adipose-derived stromal cells (ASCs) or cells of the stromal vascular fraction (SVF) is suggested to enhance the stability of the implanted fat graft. In order to improve the stability of fat tissue injection laryngoplasty using CAL, cell-seeded fat grafts with different concentrations of ASCs were analyzed in an in vivo model.

    From the adipose tissue of the neck of 14 New Zealand White Rabbits ASCs were isolated and cultivated in vitro. In a further step adipose tissue was harvested, minced and mixed with three different concentrations of labelled ASCs. Subsequently the cell-lipografts were injected subcutaneously into the auricle of the rabbits. Adipose tissue without cells served as control. After 1, 3 and 12 weeks the implants were harvested. The volume of the implants was determined and the tissue was analyzed using histological and immunhistochemical techniques.

    During 3 months after implantation there was a distinct volume reduction in all animals without significant differences between the treatment groups. Especially the constructs containing high ASC concentrations showed more areas of necrosis in the histological examinations. This results is also reflected by the immunhistochemical staining for perilipin, which revealed reduced areas of intact adipocytes during the time course.

    In this study we did not find evidence for an advantage of CAL regarding the long-term stability of the fat grafts' volume. This may be the result of an extensive processing of the injected adipose tissue.
