Synergistic tumor-suppressive effects of the flavonoid Morin and Celecoxib in head and neck squamous cell carcinoma cells
Introduction: Natural substances with potential health-promoting effects are labelled as bioceuticals. One example is the flavonoid Morin, which is contained in a variety of foods such as almonds, figs and apples. Due to its antioxidant properties, Morin has anti-inflammatory, cardioprotective and anti-carcinogenic effects, including the inhibition of cyclooxygenase (COX)-2.
Methods: The human squamous cell carcinoma cell line FaDu was treated separately and in combination with Morin and the selective COX-2 inhibitor Celecoxib. Cytotoxicity, induction of apoptosis, proliferation and autophagy were investigated by MTT test, flow cytometry and Western blot.
Results: Morin and Celecoxib promoted a reduction in cell vitality, an inhibition of tumor cell proliferation and an increased apoptosis and necrosis in FaDu cells, respectively. These effects were synergistically enhanced by combined treatment of the cells with both substances. The induction of autophagy was confirmed by the detection of an increased expression of LC3 and Beclin-1 in Western blot.
Discussion: In the present study, Morin and Celecoxib each showed tumor-suppressive effects, which were synergistic in the combination of both substances. As a mechanism of action, COX-2-induced autophagy seems to play a major role. Further studies are necessary, especially for a better understanding of the mechanisms of Morin-mediated tumor-suppression.
