Das 26S-Proteasom spielt eine zentrale Rolle in der Regulierung von Proteinen, die die Prozesse der Zellzykluskontrolle, Zelldifferenzierung, Zellproliferation und Apoptose kontrollieren. Für die Entdeckung dieses Mechanismus erhielten Aaron Ciechanover und Avram Hershko (Israel) im Jahr 2004 den Chemie-Nobelpreis. Das 26S-Proteasom, das aus einer katalytischen 20S- und zwei regulatorischen 19S-Untereinheiten besteht, ist im Nukleus und Zytoplasma aller eukaryotischen Zellen zu finden. Die 20S-Untereinheit hat proteolytische Eigenschaften, die u. a. den Enzymen Chymotrypsin und Trypsin ähneln. Substrate des Proteasoms sind Signalmoleküle, Tumorsuppressoren, Zellzyklusregulatoren, Transkriptionsfaktoren, inhibitorische Moleküle und antiapoptotische Proteine. Wenn der Abbau dieser Proteine unterbrochen wird, hat dies besonders auf sich schnell teilende Krebszellen einen Einfluss, da diese einen erhöhten Bedarf an wachstumsfördernden Proteinen haben, um die beschleunigte und unkontrollierte Mitose aufrechterhalten zu können, die ihr abnormales Zellwachstum charakterisiert. Die Erkenntnis, dass maligne Zellen offensichtlich anfälliger für den Effekt einer Proteasominhibition sind als normale Zellen, hat dazu geführt, dass dieses Prinzip Einzug in die moderne Krebstherapie gefunden hat [1] [2].

Bortezomib ist der erste Vertreter der Wirkstoffklasse der Proteasominhibitoren. Chemisch handelt es sich bei Bortezomib um ein Borsäuredipeptid, bestehend aus den beiden Aminosäuren Phenylalanin und Leucin (Abb. [1]) [3]. Die Borsäurestruktur ist verantwortlich für die Bindung an das Proteasom [4].
