LC-DAD-ESI-MS/MS, total phenolic content, and antioxidant activity of Firmiana simplex

The total phenolic content and the antioxidant activity of different extracts from Firmiana simplex (L.) fruits were investigated using Folin-Ciocalteu's reagent, 2,2'-diphenyl-1-picrylhydrazyl radical (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) and ferric reducing antioxidant power (FRAP) assays [1 – 3]. LC-DAD-ESI-MS/MS (positive and negative ionization) was used to identify the chemical composition of the most active extract [4]. Ethyl acetate (EtOAc) fraction demonstrated the highest phenolic content with 314.61, followed by 297.51, 153.75, 101.47, 97.19 for dichloromethane (DCM), butanol (BuOH), methanol (MeOH) and water (H2O), respectively, while the lowest value of 27.43 (mg GAE/g dry extract) was obtained for the petroleum ether fraction. The antioxidant activity of extracts correlated with their total phenolic content. IC50 values of the EtOAc, DCM, BuOH, MeOH, H2O and petroleum ether were 6.79, 19.92, 24.78, 28.87, 49.03 and 95.21 µg/ml, respectively, for the DPPH assay, compared to 2.92 of the positive standard ascorbic acid, whereas those of the ABTS assay were 2.11, 3.93, 6.38, 6.68, 13.27 and 40.84 µg/ml, respectively, compared to 1.63 of the positive standard trolox. IC50 values of the FRAP assay were 32.1, 26.62, 17.7, 13.35, 8.24 and 4.12 mM FeSO4 equivalent/mg dry extract, compared to 21.45 of the positive standard quercetin. LC-DAD-ESI-MS/MS of the EtOAc extract revealed the presence of phenolic acids (benzoic acid and cinnamic acid derivatives), flavonoids and tannins. This is the first phytochemical profiling of Firmiana simplex by LC-DAD-ESI-MS/MS.
