Zusammenfassung

Hochdurchsatzverfahren des „Next Generation Sequencing“ (NGS) erlauben es, kostengünstig die Nukleotidsequenzen von Millionen von RNA-Molekülen in einer Probe zu entschlüsseln. Dies ermöglicht, die Anzahl unterschiedlicher RNA-Moleküle in Gewebeverbänden oder Einzelzellen zu bestimmen und daraus Rückschlüsse zu ziehen. Dabei gibt die Gesamtheit der RNAs, insbesondere der mRNAs (messenger RNAs) als potenzielle Vorlagen für die Proteinexpression, einen umfassenden Einblick in den Funktionszustand der untersuchten Zellen und Gewebe. Hierfür können neben Zellkulturen oder frischem Gewebe mit speziellen Methoden auch formalinfixierte und in Paraffin (FFPE) eingebettete Präparate genutzt werden. In dieser Übersicht werden die methodische Strategie und ihr Einsatz im Bereich der ophthalmologischen Histopathologie dargestellt.
