Significance

Tamiflu is a potent neuraminidase inhibitor used for the treatment of the avian H5N1 virus. The key step is a putative barium-catalyzed asymmetric Diels-Alder reaction using the ligand C. A possible transition state is K. For earlier syntheses from the same group see: J. Am. Chem. Soc. 2006, 128, 6312; Org. Lett. 2007, 9, 259; Tetrahedron Lett. 2007, 48, 1403.
