Significance

Members of the polyene macrolide family have antifungal activity. The single stereogenic center in fragment C, generated by a Lewis base-catalyzed vinylogous aldol reaction (50 mmol scale), was used to introduce 8 of the 10 stereogenic centers in RK-397 using substrate control.
