Significance

FR901512 is a potent HMG-CoA reductase inhibitor (IC50 = 1.0 nM). It is the first naturally occurring statin possessing a tetralin core. The synthesis depicted twice exploits efficient catalytic asymmetric Nozaki-Hiyama allylations.
