The knowledge of human brain, with its intricate network of billions of neurons and complex structural organization, has continued to grow in the last decades: it is the epicenter of our cognition, emotion, and consciousness, orchestrating every part of the human being's existence. However, like any complex entity, even the brain can sometimes be affected by imperfections, giving rise to a compelling and multifaceted topic of study: brain and central nervous system malformations.

In this special issue, we embark on a complex journey into the captivating compartment of brain malformations, a subject that occupies a prominent place at the intersection of neurobiology, clinical medicine, and human biology. This issue is a tribute to the relentless pursuit of knowledge and the enduring commitment of researchers, clinicians, and families who confront the challenges posed by these congenital anomalies head-on.

Brain malformations encompass a wide spectrum of conditions, from neural tube defects that affect the very earliest stages of development to subtle cortical abnormalities that may not manifest until later in life.
