Acid-Mediated Synthesis of Chromeno[2,3-b]pyridine Derivatives via Condensation of 2-Amino-3-formylchromones and 1-(Methylthio)-2-nitroenamine Derivatives

Abstract

A convenient and effective protocol has been proposed for the synthesis of chromeno[2,3-b]pyridine derivatives via the CF3SO3H-catalyzed annulation of substituted 2-amino-3-formylchromones and various 1-(methylthio)-2-nitroenamine derivatives. This method is time-efficient, convenient, high-yielding, and exhibits a good functional group compatibility.

  • 18 A mixture of substituted 4-oxo-4H-1-benzopyran-3-carbonitriles (10 mmol) and concentrated NH4OH solution (10 mL) was stirred at room temperature until the solid dissolved and then was diluted with water (10 mL). The product obtained was filtered and recrystallized from ethanol to give 1 as yellow crystals
  • 19 For Aliphatic AminesTo the solution of compound 1,1-bis(methylthio)-2-nitroethylene (BMTNE, 15 mmol) in acetonitrile (40 mL), aliphatic amine (15 mmol) was added dropwise at 0 °C. The resulting solution was stirred at room temperature for 24 h. The reaction was evaporated to dryness, and the residue was purified by chromatography over silica gel to give the corresponding product.For Benzylamines and Heterocyclic AminesCorresponding amine (15 mmol) was added to the solution of compound BMTNE (15 mmol) in ethanol (40 mL). The resulting solution was stirred under reflux for 12 h. The reaction was evaporated to dryness, and the residue was purified by chromatography over silica gel to give the corresponding product.
