Significance

(-)-Englerins A and B are guaiane sesquiterpenes that are active against renal cancer cell growth. Englerin B is less active and selective than englerin A. This synthesis was published back-to-back with Ma and co-workers who also used a gold-catalyzed cycloisomerization (see previous Synfact). Overall, englerin A was synthesized in 18 steps in 7% yield.
