5-methoxylated flavones from Vochysia divergens a flood-adapted species from Brazilian Pantanal

Vochysia divergens Pohl (Vochysiaceae), commonly known as Cambará, is native in the Amazon Basin and it has considered an invasive species in the wetlands of the Brazilian Pantanal. In this work, ethanolic extract of V. divergens leaves was chemically investigated and evaluated in vitro against Schistosoma mansoni adult worms for viability and motor activity. The flavones 3′,5-dimethoxy luteolin-7-O-β-glucopyranoside (1), 5-methoxy luteolin (2) and 3′,5-dimethoxy luteolin (3) were isolated. The ethanolic extract exhibited significant reduction in motor activity: 100% for female and male adult worms of S. mansoni at 50 µg/mL after 24h. The flavones 1 and 3 were able to slightly reduce the motor activity at 25% in 24h at 25µM and 12.5µM, respectively only for male worms. The flavone 1 was identified as the major constituent from the crude extract, and 3 was not cytotoxic in the cell line V 79 (Chinese hamster lung fibroblasts) at concentrations ranging from 1.21 to 625 µg/mL. The structural elucidation was established by NMR spectroscopic methods and to the best of our knowledge 2 and 3 are reported for the first time as constituents of V. divergens.
