Abstract

An efficient synthesis of the highly stereogenic centered C10–C18 fragment of iriomoteolide-3a has been accomplished. Key steps include Sharpless asymmetric dihydroxylation and epoxidation for generation of the desired stereocenters.

10 Compound 13 was converted into a known compound 13a (Scheme 3). A comparison of specific rotations {observed for 13a: [α]_D ³⁰ +16.3 (c 1.0, CHCl₃), Lit.³: [α]_D ³⁰ +16.6 (c 1.0, CHCl₃)} showed that the enantiomeric excess in the Sharpless dihydroxylation was >95%, and also confirmed the exclusive formation of 13 from 12 (by NOE studies on 13a ¹²).
