Significance

Calcitonin gene-related peptide (CGRP) is a 37 amino acid neuropeptide found in the CNS periphery that is implicated in migraine. Interest in CGRB antagonism stems from the fact that it lacks a vasoconstrictive side effect. MK-0974 is a potent orally available CGRG receptor antagonist based on an aminocaprolactam template in phase II clinical trials for the treatment of migraine.
