The synthesis of t-butyl thymidyl ketone 13 has been accomplished starting from the substituted thymidine 9. Ketone 13 is a useful building block for the preparation of modified oligonucleotides. Irradiation of oligonucleotide 14a led selectively to cleavage products 15a and 16a.
